2.3 Model Replication Evaluation
2.3.1 General Condition
Macroscopic Changes: Starting from day 6, some mice in the aplastic anemia model group show scattered and shedding fur, especially on the neck and back, with dull fur color; food and water intake decrease compared to before, weight drops, spirits are poor, activity is less flexible, and reactions are sluggish.
2.3.2 Peripheral Blood Count
One week after modeling, 20 μl of blood is collected from the tail vein of each mouse (fasting for 12 hours before blood collection, but allowed to drink water) and mixed with 20 μl of anticoagulant in a test tube, then sent to the laboratory for routine blood tests. The results show significant differences between the normal group and the model group in Hb, WBC, and PLT (P < 0.05), while RBC in the normal group is also lower than in the model group, but the difference is not significant. 2.3.3 Bone Marrow Smear
One week later, 5 mice are randomly selected from each group. After decapitation, the left femur is removed, and 1 ml of RPMI-1640 solution is injected through a No. 4 needle to flush out the bone marrow onto a slide, followed by HE staining and microscopic observation of the bone marrow smear. The results show that the bone marrow smear of the model mice exhibits hypoplasia, with the granulocytic lineage averaging less than 15%, the erythroid lineage averaging less than 7%, and 0–9 megakaryocytes. In contrast, the normal group shows active bone marrow proliferation, with the granulocytic lineage averaging more than 40%, the erythroid lineage averaging more than 14%, and 45–72 megakaryocytes. There is a significant difference between the two groups.
2.3.4 Model Evaluation
After modeling, the general condition of the mice is poor, with decreased peripheral blood counts and bone marrow parameters, showing significant differences compared with the normal group (p < 0.05 or p < 0.01), indicating that the model replication was successful and subsequent experiments can proceed.
