2.5 Ultra-thin Section Electron Microscopy Observation
Randomly take 0.1cm³ of tumor tissue from each group, quickly immerse the tissue in 2.5% glutaraldehyde fixative for more than 2 hours, rinse with phosphate buffer, fix with 1% osmium acid for 1–2 hours, rinse again with buffer for 20 minutes, dehydrate using acetone gradient, impregnate, embed, prepare ultra-thin sections, stain with uranyl acetate and lead citrate, observe under transmission electron microscope, and take photographs [22–23].
