2.5.1 Mouse Bone Marrow Nucleated Cell (BMNC) Count
The hemocytometer and coverslip are cleaned, and the coverslip is placed on the hemocytometer. A small amount of cell suspension is drawn out and dropped on the edge of the coverslip so that the suspension fills the space between the coverslip and the hemocytometer, and left to sit for 3 minutes. Under the microscope, the total number of cells in the four large squares of the hemocytometer is counted, with cells crossing the lines only counted on the left and top sides. Then the calculation is as follows:
Cell count/ml = (Total number of cells in 4 large squares / 4) × 10000
