2.5.2 Bone Marrow Tissue Morphological Observation

The sections are observed under high magnification (10×40) with an optical microscope. Following the method described in literature [29], a 5×5 grid micrometer is used to measure the bone marrow hematopoietic tissue volume (v%) by point-counting method. Ten random fields are selected, and the targets hit in each field are recorded. If the intersection hits hematopoietic tissue, fat tissue, or blood sinus, it is recorded as 1 point; if it hits the boundary between hematopoietic/fat tissue, it is recorded as 1/2 point. A total of 100 points are observed, and the percentage of hematopoietic tissue, fat tissue, and blood sinus is calculated as follows:

Hematopoietic tissue vol% = (Number of hits in hematopoietic tissue / (Number of hits in hematopoietic tissue + Number of hits in fat tissue + Number of hits in blood sinus)) × 100%

Fat tissue vol% = (Number of hits in fat tissue / (Number of hits in hematopoietic tissue + Number of hits in fat tissue + Number of hits in blood sinus)) × 100%
