Chinese Abstract

Objective: To investigate the detoxifying and efficacy-enhancing effects of Pei’s Shengxue Granules (PG) combined with 5-Fu on tumor-bearing mice by observing their impact on bone marrow DNA content and cytokine secretion (TNF-α and IFN-γ).

Methods: A tumor-bearing H<sub>22</sub> (hepatocellular carcinoma) mouse model was established. Mice were divided into a normal group, a model group, a 5-Fu group, and PG high-, medium-, and low-dose + 5-Fu groups. After 10 days of administration, tumor inhibition rates were calculated, peripheral blood and bone marrow DNA contents were measured, serum TNF-α concentrations were determined using radioimmunoassay, and serum IFN-γ concentrations were measured using ELISA.

Results:

  1. The tumor weights in the high-, medium-, and low-dose PG + 5-Fu groups were all lower than those in the 5-Fu group, with the medium-dose PG + 5-Fu group showing the most significant reduction (p<0.05), achieving a tumor inhibition rate of 61.07%.
  2. The WBC and PLT counts in the high- and medium-dose PG + 5-Fu groups were significantly higher than those in the 5-Fu group, with statistically significant differences (p<0.05).
  3. The bone marrow DNA content in the high-, medium-, and low-dose PG + 5-Fu groups was all significantly higher than that in the 5-Fu group, with the high- and medium-dose groups showing particularly significant differences (p<0.05).
  4. The serum TNF-α concentrations in the high-, medium-, and low-dose PG + 5-Fu groups were all higher than those in the 5-Fu group, with the high-dose PG + 5-Fu group reaching 0.71 ng/mL, which was significantly different from the 5-Fu group (p<0.05).
  5. The serum IFN-γ concentrations in the high-, medium-, and low-dose PG + 5-Fu groups were all significantly higher than those in the 5-Fu group, with the high- and medium-dose groups at 53.7 pg/mL and 56.9 pg/mL respectively, both showing significant differences (p<0.05).

Conclusion: Pei’s Shengxue Granules can significantly increase the tumor inhibition rate of 5-Fu, enhance the bone marrow hematopoietic function of chemotherapy-treated mice, and promote the secretion of cytokines TNF-α and IFN-γ, indicating that Pei’s Shengxue Granules have a detoxifying and efficacy-enhancing effect on 5-Fu chemotherapy in tumor-bearing mice.
