The Effect of Pei's Hematopoietic Granules on Serum EGF and IL-1 Levels in H₂₂ Tumor-Bearing Mice

Chinese Abstract
Objective: By observing the effect of Pei's Hematopoietic Granules (PG) on serum Epidermal Growth Factor (EGF) and Interleukin-1 (IL-1) levels in H₂₂ tumor-bearing mice, this study explores the regulatory role of PG on the immune system of tumor-bearing mice as well as its inhibitory effect on tumors.
Methods: A H₂₂ tumor-bearing mouse model was established by subcutaneous injection at the right anterior axillary region. The inhibitory effect of PG on mouse tumors was observed, and the concentrations of Epidermal Growth Factor (EGF) in mouse serum were measured using ELISA, while the concentrations of Interleukin-1 (IL-1) in mouse serum were detected using radioimmunoassay.
Results:
- The average tumor weights in the high-, medium-, and low-dose PG groups were all lower than those in the model group, at 1.708±0.396 g, 1.623±0.365 g, and 1.808±0.462 g, respectively, with statistically significant differences compared to the model group (p<0.05). The tumor inhibition rates were 32.1%, 35.5%, and 28.1%, respectively.
- The thymus index (TI) and spleen index (SI) in the high-, medium-, and low-dose PG groups were all higher than those in the model group. Specifically, the TI values for the medium- and high-dose groups were 38.7±9.8 mg/10 g and 36.5±9.2 mg/10 g, representing increases of 37.2% and 29.2% compared to the model group, both with statistically significant differences (p<0.05). The SI value for the medium-dose group was 70.5±19.0 mg/10 g, a 29.9% increase compared to the model group, also with statistically significant differences (p<0.05).
- The EGF concentrations in the high-, medium-, and low-dose PG groups were all lower than those in the model group, with the medium-dose group showing an EGF concentration of 6.377±2.414 ng/ml, which was statistically significantly lower than that of the model group (p<0.05).
- The IL-1 concentrations in the high-, medium-, and low-dose PG groups were all lower than those in the model group, with the medium- and high-dose groups having IL-1 concentrations of 0.099±0.028 ng/ml and 0.078±0.027 ng/ml, respectively, both showing statistically significant differences compared to the model group (p<0.05).
Conclusion: Pei's Hematopoietic Granules can increase the weight of the thymus and spleen in H₂₂ tumor-bearing mice, inhibit the production of EGF and the secretion of IL-1, thereby exerting an anti-tumor effect.
