2.2 Animal Grouping and Dosage
2.2.1 Grouping
Seventy-two SPF-grade Kunming mice are randomly divided into 12 as the blank control group, and the remaining 60 are weighed 24 hours after inoculation and then randomly assigned into 5 groups of 12 each, namely: tumor-bearing control group, Pei’s Soft Liver and Anti-Bloating Pill (PRGXP) large, medium, and small dose groups, and Compound Cantharidin Capsules group (BM).
2.2.2 Administration and Dosage
Pei’s Soft Liver and Anti-Bloating Pill and Compound Cantharidin Capsules are finely ground before administration and made into suspensions with distilled water. According to the gastric volume of mice, 0.2 ml/10 g is administered.
Blank Control Group: 0.4 ml of distilled water per mouse.
Compound Cantharidin Capsules Group: Each capsule contains 0.25 g, adults take 2 times daily, 3 capsules each time. Assuming an adult weighs 60 kg, the daily dosage for adults is 0.025 g/kg·d, while the dosage for mice is equivalent to 10 times the adult dosage, i.e., 0.25 g/kg·d.
Pei’s Soft Liver and Anti-Bloating Pill Group: 6 g per packet, adults take 2 times daily, 1 packet each time. Assuming an adult weighs 60 kg, the daily dosage for adults is 0.2 g/kg·d, while the low-dose for mice is equivalent to 5 times the adult dosage, i.e., 1 g/kg; the medium-dose for mice is equivalent to 10 times the adult dosage, i.e., 2 g/kg; the high-dose for mice is equivalent to 20 times the adult dosage, i.e., 4 g/kg. Each group receives medication once daily at 8–9 am, dosed according to weight, for a total of 12 days, after which the mice are euthanized by decapitation.
