Chinese Abstract

Objective: To explore the regulatory effect of Peishishengxue Granule (PG) on the immune system of H₂₂-bearing mice by observing its impact on the weight of immune organs (thymus and spleen), the proliferation activity of splenic lymphocytes, and the secretion of cytokines (IL-2 and IFN-γ).

Methods: A solid tumor model was established by implanting H₂₂ hepatocarcinoma cells into mice. The weights of the thymus and spleen were measured at the end of the experiment, and the thymus index (TI) and spleen index (SI) were calculated. Lymphocyte proliferation was assessed using the MTT colorimetric assay, while the concentration of interleukin-2 (IL-2) in the supernatant of splenocyte cultures was determined by enzyme-linked immunosorbent assay (ELISA). The mRNA levels of IL-2 and IFN-γ in splenocytes were quantified using reverse transcription-polymerase chain reaction (RT-PCR).

Results: In all PG dosage groups, the TI and SI of the H₂₂-bearing mice were higher than those of the model group. Specifically, in the high-dose group, the TI and SI were 18.9 mg/10 g and 76.3 mg/10 g, respectively, representing increases of 36.0% and 17.9% compared with the model group (p<0.05). In the medium-dose group, the TI was 18.0 mg/10 g, a 29.5% increase over the model group (p<0.05). Furthermore, the proliferation activity of splenic lymphocytes in all PG dosage groups was higher than that in the model group, with the high-dose group showing the most significant enhancement (p<0.05). The concentration of IL-2 in the supernatants of splenocyte cultures was also elevated in all PG dosage groups compared with the model group, reaching 61.6 pg/ml in the high-dose group, which was significantly higher than the model group (p<0.05). Additionally, the mRNA levels of IL-2 and IFN-γ in splenocytes were increased in all PG dosage groups, with significant differences observed in the medium- and high-dose groups compared with the model group (p<0.05).

Conclusion: Peishishengxue Granule has the effect of increasing the weight of immune organs (thymus and spleen) in H₂₂-bearing mice, enhancing the proliferation activity of splenic lymphocytes, promoting the secretion of the cytokine IL-2, and upregulating the expression of IL-2 mRNA and IFN-γ mRNA. These findings indicate that Peishishengxue Granule can significantly enhance the immune function of H₂₂-bearing mice and inhibit tumor growth.
