2.6.3 Immunohistochemical Index Detection

The positive reaction product of P27 protein is mainly located in the nucleus and cytoplasm, with brown-yellow or brown-brown granules in the nucleus and cytoplasm indicating positive expression. The positive reaction product of Bcl-2 protein is mainly located in the cytoplasm, with brown-yellow or brown-brown staining granules in the cytoplasm indicating positive expression. Under a 400-fold optical microscope, randomly select 5 sections from each group and randomly sample 3 fields of view from each section, totaling 15 fields of view per group. Use the BI-2000 medical image analysis software to detect the average optical density (MOD) of the stained positive substances for P27 and Bcl-2, which respectively represent the content of P27 and Bcl-2. Average optical density (MOD) = Integral Optical Density (IOD) / Area (A). The higher the MOD value, the greater the amount of the measured substance or the deeper the color after staining, indicating a larger quantity.

2.7 Statistical Methods

Use SPSS 17.0 statistical software for data processing. All experimental values are expressed as mean ± standard deviation (x±s). One-way ANOVA is used for comparing multiple means. A p-value less than 0.05 is considered statistically significant.
