Conclusion
Pei’s Shengxue Granules can significantly improve the tumor inhibition rate of 5-Fu, enhance the bone marrow hematopoietic function of mice after chemotherapy, and promote the secretion of cytokines TNF-α and IFN-γ, demonstrating a detoxifying and efficacy-enhancing effect on 5-Fu chemotherapy in tumor-bearing mice.

Foreword
Pei’s Shengxue Granules is a specialized prescription formulated by Professor Pei Zhengxue, a renowned expert in integrated traditional Chinese and Western medicine, for the treatment of leukemia [1]. It mainly consists of Liuwei Dihuang Tang combined with Shengmai San, supplemented with Taizishen, Beishashen, Dangshen, and other herbs, with emphasis on strengthening the spleen through Taizishen, Dangshen, and Beishashen, and nourishing the kidneys with Liuwei Dihuang Tang [2][3][10]. This formula gained fame after completely curing a patient with acute monocytic leukemia (M₅), Ma Changsheng, and was officially named the “Lanzhou Formula” at the Hematology Conference held in Suzhou in 1974. In 2003, Pei’s Shengxue Granules successfully cured another leukemia patient (L₂), Liu Ligang, as reported in the Nanchang Daily and the Gansu Daily. In 1997, the formula was developed into an internal preparation called “Pei’s Shengxue Granules” by the Gansu Provincial Academy of Medical Sciences. Starting in 2002, this formulation was used as a clinical research project at the Gansu Academy of Medical Sciences, passed the evaluation of results, and won the First Prize of the Huangfu Mi Science and Technology Achievement Award in Gansu Province in 2003 [118].

Over 30 years of clinical application have proven that Pei’s Shengxue Granules (also known as the “Lanzhou Formula”) not only show excellent efficacy against leukemia but also effectively improve immune dysfunction caused by radiotherapy and chemotherapy in malignant tumors. Clinical practice has demonstrated that this formula is highly effective against esophageal cancer, gastric cancer, liver cancer, malignant lymphoma, and aplastic anemia. According to incomplete statistics during my time assisting in clinical practice, it has cured three cases of esophageal cancer, three cases of leukemia, three cases of aplastic anemia, one case of gastric cancer, and three cases of primary liver cancer. Xue [4] used the “Lanzhou Formula” to treat primary liver cancer, resulting in a significant reduction in tumor size, complete recovery of liver function, improved quality of life, and prolonged survival; Li et al. [5] conducted an efficacy observation on 10 cancer patients treated with the Lanzhou Formula in combination with chemotherapy, showing that the short-term efficacy of the Lanzhou Formula group was superior to that of the chemotherapy-only group (p<0.05), while the long-term side effects were significantly milder in the Lanzhou Formula group (p<0.01). When Pei’s Shengxue Granules were used in conjunction with radionuclide therapy for 15 cases of liver cancer, with the treatment group receiving radionuclide therapy plus PG and the control group receiving radionuclide therapy alone, the treatment group exhibited markedly fewer side effects than the control group, and the therapeutic effects of the two groups also differed significantly [6].
In terms of experimental research, previous studies have explored the anti-tumor mechanisms of Pei’s Shengxue Granules from perspectives such as regulating the body’s immunity, inducing apoptosis, inhibiting mutant p53, and upregulating Caspase-3 protein expression [7–10], but research on the chemotherapy-detoxifying and efficacy-enhancing effects of Pei’s Shengxue Granules remains largely unexplored. This experiment aims to fill that gap by conducting a study and discussion on this issue.
This experiment establishes a tumor-bearing H22 mouse model and administers 5-Fu chemotherapy, observing the efficacy-enhancing and detoxifying effects of Pei’s Shengxue Granules on chemotherapy in tumor-bearing mice. To explore the mechanisms behind Pei’s Shengxue Granules’ detoxifying and efficacy-enhancing effects on H22 tumor-bearing mice, this study focuses on improving bone marrow suppression and regulating immunity, aiming to provide scientific and microscopic evidence for the widespread clinical application of Pei’s Shengxue Granules.
